TA cloning is also known as rapid cloning. It is a sub cloning technique. It can be performed without the use of restriction enzymes.

It is an easy and simple method of cloning compared to the traditional sub cloning.

TA cloning is based on the Adenine Thymine base pairing on different DNA fragments to hybridize.

PCR product is generated using Taq Polymerase, which adds A overhangs to the 3’end of the product.

PCR product with the A overhang can be ligated to linearized vector having T overhang
